FGF Signalling in Vertebrate Development

The fibroblast growth factors FGFs represent one of the relatively few families of extracellular signalling peptides that have been shown in recent decades to be key regulators of metazoan development. FGFs are required for multiple processes in both protostome and deuterostome groups. Given the wide range of regulatory roles attributed to the FGFs, it is perhaps not surprising that misregulation of this signalling pathway has been implicated in a number of human disease conditions.

The focus of the present review is to look at the fundamental components of the FGF pathway and illustrate how this highly conserved regulatory cassette has been deployed to regulate multiple, diverse processes during vertebrate development.

Fibroblast growth factor 2 FGF2 is one of the most studied growth factors to date. Most attention has been dedicated to the smallest, 18kDa FGF2 variant that is released by cells and acts through activation of cell-surface FGF-receptor tyrosine kinases. In this review, we summarize the current knowledge about the biology of HMW FGF2, its role in disease and areas for future investigation. FGF2 is a growth factor that exists in several isoforms differing in their N-terminal extensions, subcellular distribution and function.

This review will focus specifically on nuclear HMW FGF2 signalling, its molecular mechanisms and its physiological and pathological function. FGF-8 belongs to the fibroblast growth factor FGF family and plays an important role in cell growth, embryogenesis, and tumorigenesis. The isoforms may have different functions during embryogenesis. During prenatal stages, FGF-8 is required for the normal development of various organs, including limbs and central nervous system. FGF-8 was first cloned from SC-3 mouse mammary carcinoma cells and was found to be induced in response to androgenic stimuli.
